Drug Repurposing Candidates for Triple-Negative Breast Cancer

Discover (after iteration 4) — find something NEW that iteration 4 did not establish: a pattern, formula, conjecture or effect that no published work reports. Start from what is still open: Fix the drug-name matching (use Drug ID or the correct name column), then preregister the mk-1775 max-T and empirical-Bayes test with the lines frozen; Count PRISM/CTRP drug overlap, using nutlin-3a as the positive control; Test b(n) on independent drugs and cell lines. Derive the t-theory prediction to check whether the scaling is merely a restatement of it; Extend the exact enumeration to larger n and try to derive or disprove the parity pattern; Wet-lab adavosertib validation across the 9 TNBC vs 9 non-TNBC lines. Compute many exact small cases, state one precise conjecture, test it on cases not used to find it, try to break it with counterexamples, and check its novelty against the literature.

  1. Iteration 5 · Exact theory of leave-one-out sign-flip parity and sqrt(n-1) scaling in small-n drug screens, plus fixed mk-1775 test

    Completed
    • Agent 1 · lead

Builds on the jackknife sign-flip law: derive the analytic t-theory prediction of b(n) (delta method / noncentral t) and test whether sqrt(n-1) is a restatement; validate on independent drugs and cell lines (non-breast lineages, PRISM/CTRP where available) with multiple seeds and report the exponent distribution.

    • Agent 2 · explorer

Pattern hunter: Builds on exact n=3..8 enumeration: extend with DP or generating functions to n>=14 (including ties and unequal group sizes), state one precise parity conjecture, test it on held-out n, search for counterexamples and check OEIS and the literature.

    • Agent 3 · experimenter

Skeptic: fix drug-name matching by Drug ID with asserted match counts, run the frozen 9v9 mk-1775 max-T and empirical-Bayes test, count PRISM/CTRP drug overlap using nutlin-3a as the positive control, and attack the other agents' claims for bugs and known results.

    • Agent 4 · reviewer

Independent review of claim c5a1ed80-5b2b-4085-a6b5-c6302d90cab4: Rank-sum LOO sign-flip parity: alternation persists for equal groups n=3..12, absent for m≠n (exploratory)

    • Agent 5 · reviewer

      Independent review of claim c64d7aa6-77d2-4cf7-9358-1dfc54355380: mk-1775 9v9 within-breast GDSC2 by DRUG_ID: max-T p=0.040, moderated-t q=0.009 (exploratory)
